Can i adjust the float level (screw and nut) with the engine turned off?

No because you don't have an fuel pressure. The needle and seat you are adjusting works against the fuel pressure. You also won't have anwhere for the fuel to go if you lower float level--it won't get sucked in a stopped engine.
