I would connect the gauge, disconnect the vacuum hose and plug it, start the car, adjust the pressure to the 43.5 or whatever, shut off the engine, then disconnect the gauge.

Not sure why you're adjusting fuel pressure after you've removed the gauge...how do you know what your adjusting it to w/ no gauge attached?

1.) Attach gauge.
2.) Disconnect and cap vacuum.
3.) Start car.
4.) Adjust fuel pressure.
5.) Shut off car.
6.) Remove gauge.
7.) Reconnect vacuum.

If you adjust the fuel pressure to 45psi w/ no vacuum connected to the regulator, it should drop to ~36 or so PSI when you re-connect the vacuum to the fuel pressure regulator...depending on how much vacuum your engine pulls at idle.
