What would the difference be? How much less mileage and how much will the speedo be off by? This is on an open differential 3.1 V6. Would it be worth it?

Absolutely not worth the time, money, and effort, especially with a V6. The ratio difference is negligible, and it's not going to make the V6 grow any more power.
