VE and AFPR tuning

Engine: 383 single plane efi
Transmission: T-56
Axle/Gears: 8.8 with 3.73s
VE and AFPR tuning
I have been gong on about tuning my new engine, but I have come to the question of how to tune it with the AFPR? I can change things like VE to get the right ratio I want and then turn the AFPR up or down and do it agian and do that all day long. How though do you go about doing it. Is it a day at the dyno event or six months in the making ordeal?
