Adjust fuel pressure w/ vacuum line on or off?

Is the fuel pressure supposed to be adjusted w/ the vacuum line connected to the hat? or disconnected but plugged? And with the vacuum line connected should the pressure be about 42 PSI or is that with the vacuum line off?


I presume turning the screw clockwise will increase the pressure, correct?

Disconnected, hose plugged.
