The S truck transmission is not rotated like it is in our cars; and the shifter is in a different spot from front to rear (farther forward IIRC). The transmissions for the 2 chassis do not interchange. Lots of internal parts do though; but not all.
A 6-cyl one has a small-diameter clutch gear with 14 splines for the clutch disc; a V8 one has a much larger gear with 26 splines.
The 6-cyl transmissions have performance-hostile gear ratios; very wide spacing between the gears. Very low 1st gear, to the point that it's totally useless with a motor with any appreciable torque; on the other hand, if you gear the rear high enough to where 1st works, all the other gears (3rd on up especially) are too high. V8 transmissions have much better ratios, although still wider spaced than ideal.

ok, the main reason i posted this is i am buying a thirdgen camaro. here is where it gets tricky. i spent the last 2 years building up and making gobs of power out of the l67 3.8 v6. i want to put one in a camaro with a 5speed. i kept hearing that it wont line up with a rwd trans. i know a 3.8 come in a 98+ camaro and has a stick. i also know the 3.8 will go directly in with a 2.8 fiero with no mods to the housing. so if the v8 t-5 was a different housing patter, then i could get a 2.8 camaro 5spd and rebuild the t-5 with some heavy duty parts to slip right on the 3.8. i think i made some sence there but im not sure.

I have no clue what bolt pattern the 3.8 has. I know for certain that a V8 bell housing will NOT bolt up to a 60° V6, or vice-versa.
But if the 3.8 and the 2.8 are both 60° 6-cyls, then there's a possibility that it will work.
I know nothing about them myself though; I've never so much as touched a 6-cyl motor in one of these cars, can't remember that I've ever even looked at one except accidentally. So I can't help you much more than that.
