From: Littleton, CO USA
Car: 82 Berlinetta/57 Bel Air
Engine: L92/LQ4 (both w/4" stroke)
Transmission: 4L80E/4L80E
Axle/Gears: 12B-3.73/9"-3.89
Difference:
Roller lifters have a roller on the bottom that follows the cam lobe. Need some means of keeping the lifter from rotating in its bore, and the cam from walking forward and aft in the engine. More expensive than flat.
Flat lifters are flat on the bottom. They have a hardened face that follows the cam lobe. Less expensive than roller set ups.
The cam and lifter type have to be meant for each other.

Benefits:
Flat are less expensive.
Roller can have a more aggressive profile to get more air/fuel in the cylinder and exhaust out.

Did I mention that roller lifters and cams are more expensive than flat lifters and cams?
