Valve adjustment hydraulic roller
I built a vortec 350 for the 85 iroc. I adjusted the valves with the motor out of the car. When i adjusted them, i tightened the rocker nuts while turning the pushrod until i felt some tension build then i proceeded another 1/2 turn. When i fired it up for the first time, it fired instantly and ran smooth. Do i need to readjust the valves after its supplied a sufficient amount of oil to all parts of the engine?

That 1/2 turn past zero lash is the required preload. That's all the adjustment they require. If they need additional adjustment, something is wrong.
