Setting Timeing

Ok I just rebuild the 350 in my gta and i have it all together in the car and it runs but I can't seem to get the timming right. It gets close and sounds good then it gets really rough and sometimes stalls out. I did disconnect the signal wire from the distributor to the computer. Is there anything else that i am missing. Is it just a vacuum leak or are they just hard to set??

you disconnected the correct wire, right?
the EST wire thats on the passenger side firewall, tan in color, ??
also did you put it in correctly? it may be 1 tooth off or 180* off.
