Engine: 305 (LO3)
Transmission: 5 spd manual
Run engine supported by jack, with transmission removed.

To make sure that a new flywheel and pressure plate is balanced properly before complete assembly.

Good idea or should some more ridged means of support to the rear of the engine be used?

Ive done it before. Is it best? Probably not. But Ive done it.

Just put a block of wood between the jack and oil pan.

Revs a lot easier!

theoretically I don't see why not, just make sure that block of wood doesn't come in contact with the flywheel. I wouldn't bounce it off the rev limiter while doing it but light revs to 2500 would seem fine.
