Has anyone ever made the factory fender vents on 3rd gen T/A's functional?

They already are functional. They vent engine compartment air out. They were designed like that from the start. Part of the aerodynamic design of the 3rd gen style.

Only the passenger side vents. The driver side is completely sealed off from the engine compartment...

I think we are have been looking at these vents the wrong way for too long. Note that the ad talks about pressure build up. Not getting heat out. I think these are made to relieve air pressure built inside the fenders, which cause drag and lift. If these are 'exhaust' vents for the air pressure build up inside the fenders, then they are indeed functional. I wanted to believe GM used them like Chrysler did with the Plymouth Superbird, but that would require the fender liner behind the wheels to be opened up. Maybe that's a mod that could be made? If done, then a block plate to isolate that lower part of the fender would be in order, perhaps? Either way, they might be functional after all, even though not in the way most of us thought they were.
