500 horse on pump gas?
Aftermarket +190cc heads, solid or roller cam, +230 duration/+0.500lift cam, 1 3/4" headers, +10:1 compression are yer basic minimums.
Best thing about 400's really is the torque. Spend at least $800 on a good set of heads and yer basically guaranteed +400hp/400lb-ft(1500-5000) with something like an XE274 camshaft which is a great street/strip cam. Very Idle friendly in a 400.
If I was making a motor for what I thought would be guaranteed 500hp I'd go with 210cc AFR, Canfield, Pro Topline, Brodix or Dart heads(CNC'd aluminum heads for sure to make it street friendly), 10.5:1 compression, rpm air gap or super victor intake, 1 3/4 headers and either hyd roller cam with 240-250 duration and about 0.550 lift(really this would depend on the intake) or a solid grind with about the same specs... really depends on alot of variables for the cam.

500hp is a pretty lofty goal. You will need awesome heads ie AFR 210's. A big solid roller cam ie XR280 or XR286. You will also need a bullet-proof bottom end, read forged everything.
