This is the motor I want to replace my 91 305TPI 5 Speed, Will this bolt right up? Will I need a new tranny since mine is a manual?

It's a GM crate motor, available from your local friendly GM dealer.
It will "bolt right up" to the chassis and transmission. That's where the good news ends.
You'll be doing an EFI to carb conversion as well. There are a lot of details involved in that. There's a tech article on it linked on the home page, although you won't be doing the intake manifold swap portion (it will already be on the engine).
The big issue is whether your T5 will stand up to the 350. It will if you treat it right, but it won't take the abuse the 350 HO is able to dish out.

If I had a TPI car and wanted the 350 HO engine, this is the way I'd go http://www.sdpc2000.com/catalog/1578...rip-Engine.htm . It's more money by the time you get it set up as intended (runners & TB, for instance), but IMHO, you'll have a better package when you're done.
Besides, the HO Deluxe has the "wrong" carb for a manual transmission, anyway.

I have been looking at that same motor for my 89 Ta, but mine is a 700r4. I think I am going to get it.

If you are to buy a crate motor from GM, say for example the 383 with 385hp, will it pass emmisions with all of the bolt ons.
Like the smog pump and stuff.

Some people who don't have strict visual inspections have been able to get them by. One person in California got one through by passing it off as something else. Still haven't heard if Chevy High Performance magazine got their Vortec-headed 305 past Herr Inspektor.
The HO doesn't have heat cross-over passages in the heads, so exhaust has to be plumbed up to the intake manifold in order for the EGR to work.
