Is there supposed to be a screw in it? I'm not sure if the previous owner did it or if it is factory. Both sides.

what do you mean by roof rail??do you mean the TTop bar
Hardtop, along the roof rail, inside trim.

Yup. No screws, just those plastic clips that James showed. I guess the PO decided to use screws instead of replacing the broken clips.
