Engine: 5.7l TPI
Transmission: 700r4
Axle/Gears: Stock
Can an intake leak cause a misfire?

My car is misfiring with a thumping noise out of the exhaust and when they ran the code they said it was a random misfire, and that both cylinder 1 and 4 are misfiring

Yes, having more air in the combustion chamber than the ECM thinks is there can definitely cause a misfire.
