TPI timing

Engine: 355TPI
Transmission: T-5
Axle/Gears: 3.27
TPI timing
So Ive gone through some threads searching for my question but didnt run into one. My question is When you put the car on timing do you disconnect the connector, set it on 4*,turn it off, reconnect the connector and turn on, OR Do it all while it's on? Does the MSD 6AL box have anything to do with it? and i also want know what to put it on 4* or 6*. most of the ones I read said 4* but just making sure, your help will be very appreciated. thanks

Thanks man, will try that. right now the car is running good but i just guessed at it didnt even use a gun just put it on timing by sound and how it ran.
