I recently Bought a rear end for my chevy and its got 3.40 gears, 10 bolt, and its a Locked differential. My question is, how much of the posi lube should i use with the gear oil and should i use 80 90 gear oil?

Locked as in spool type, actual locker, or limited slip(posi)? limited slip is the only one that needs the additive. I would go with synthetic no matter which you have.

And I assume the actual ratio is 3.42.

The normal dose of the additive is one tube for petroleum and most synthetic lubes for one fill (put the additive in, then fill to the bottom of the hole with gear lube). If you go with AMSOIL Series 2000 75W90 gear lube, you don't need the additive. Assuming this is a clutch-type limited slip carrier: If something like a PowerTrax, you again don't need the additive.
