i have a 305 tpi with 3.27 rear non posi will a posi-rearend make a BIG CHANGE or not. I am going to build a 350 tpi soon .

Changing your gear ratio to a 3:42 or 3:73 would produce a more noticeable difference than just swapping to posi. Although I don't know if the 305 has the torque needed to really take advantage of the 3:73 ratio.

If you have the 3.27 gears, it is almost certainly already a posi, as that's a 9-bolt ratio, and virtually all of those had it.
Furthermore, you already have a pretty decent gear for the RPM-limited TPI system. There is very little bang for the buck available there. WAY too much maze, WAY not enough cheeze.
IMO your greatest opportunity for improvement ATM, for the cheeeepest, lies in reversing the last 2 digits of your engine size.
