Which "big nut"? The fuel filter housing, or the fuel line going into the fuel filter housing? What kind of wrench are you using? How are you pulling on the wrench? Are you just pullllling on it? Use short/hard jolts; this will usually break something free. You can also tap the wrench with a hammer. (Make sure you're turning it counter-clockwise, of course.
) Be careful when tapping the wrench with a hammer; this can cause sparks and/or chipping of the wrench, so first make sure there's no fuel that can ignite from a spark, and secondly, wear eye protection so you don't get a metal shard of wrench in your eye.
If you're trying to get the fuel line nut out of the fuel filter housing, are you using a flare nut wrench? THis helps prevent rounding of the fuel line nut.
Once off, the fuel line will be a pain in the butt to get back into the fuel filter housing. This is because you'll bend the metal fuel line slightly when removing it. This causes the nut to go into the housing at an angle... so keep it in mind.
When you get the fuel filter housing out of the carb, you might want to use teflon tape on the threads before re-installing it to help prevent against leaks.

Referring to that 2.8 carb set up & the fuel line?
I owned my '85 S-10 Blzer since 1989.
I changed the fuel filter twice in my ownership.
Once on the car.
Next time was when I rebuilt the carb, last year, while it was on the bench.
Wait until you do a fuel pump job.
I had to cut the hard fuel line to get it back on the pump. I used rubber fuel line hose to patch the break I made.
I'd love to kick the engineer in the a$$ that designed such a lousy set up that doesn't allow easy repairs (unlike a regular V-8 carb fuel system set up).
The nut is a 1" wrench size. The small nut (NOT YOU!!), definetly need what Tom said, flare wrenches. METRIC!!!!
Good luck.
It really sucks.
Been there.
I actually had a friend take the nut off the carb. He had a larger selection of wrenches.
