I never use the stuff anywhere it can get into something that will clog like a tranny or brake system. Try to tighten it more, and if that doesn't work, check the flare to make sure it is OK.

Don't use the old style Teflon tape, it eventually breaks up and gets into the fluid. Insted, use a Teflon O-ring.

If you're talking about th fitting that goes into the radiator (NPT) then yes...

If you're talking about the flared fitting that couples the line to the fitting on the radiator, then no you should not, reason being is that you'll only be doing a bandaid fix, which will eventually leak again.

You should be able to tighten the fitting snug, then an 1/8th to a 1/4 turn more (holding the "nut" side of the fitting on the radiator) and it should seal properly, if not...look for a crack/split on the "flare" of the tranny line, or a burr on the taper of the fitting.

The tranny line is a flared line, designed to be sealed via pressure of the nut, not with tape/sealant...so if it's leaking there, look for damage and replace as needed.
