I just bought a 10 bolt with a 3.42 and ls1 disc conversion... it doesnt have the vent on the top passenger side..can I plug that hole with a rubber plug from the auto store or does it have to be vented?

It should be vented. You can pick up simple vents that prevent dirt from entering the diff.
One of these is what you need depending what size the hole is.

It you don't vent it, oil will be forced past the seals from the pressure that will build when it gets hot.

Simple and cheap solution. Just take a pair of pliers to the junkyard and yank one out of a junk rear, put it in your pocket, and take it home. They just press in and they're pretty easy to pull out with a pair of pliers.
