Axle/Gears: 4.10 D44

That's the right place. Middle of the block (front to rear), just above the oil pan rail. The knock sensor is plugged into the same hole on the passenger side.
There may be enough accumulated gunk that nothing comes out when you remove the plug. If so, poke it with a screw driver and you should get flow.

I seemed to have rounded it off while trying to remove it. Any suggestions on how to get it out?

Might help to file it down to a nice sharp fresh-looking hex. An impact socket, to go along with using impact rather than a steady pull, is always better than a "chrome" one.
A drill is the next step.

The two sides of the block are not connected (coolant-wise). The knock sensor drained that side of the block. The drivers side did not drain. But the drain/fill/drain method will dilute whatever is in there.
