Usually has a one way valve on it and possibly goes to the plugged line off the TB shown in 2nd pic
Normally has a connection to a vac reservoir somewhere as well
TPI one shown but same setup

* UPDATE *
i now know what that is , yeah it is a vacuum line that goes from driver side sensor (map or whatever sensor is that ) how i figured it out. i had check engine light on (code 34) i saw that the vac line was connected at the sensor but the other end of the line was not connected to anything so i plugged it into that tube shown in the pic. and the check light went off.

Your MAP sensor is located in your first pic, upper left corner, attached to the cowl lip. It is missing a vacuum line in your pic - maybe that's it in your last pic that's plugged off. The vacuum connector you are questioning looks like the one for the heater diverter valve.

im not sure about that some people say that there should be no vacuum line hooked up to that sensor on L69 HO engines but should be hooked up to the other sensor on driver side. that sensor looks the same as on the pass side but has gray connector instead of green connector. i will take pics tomorrow to show what im talking about.
here is a link i came across when i did research

The vacuume line from the map goes to the back of the tb, in your second pic it has a plug in it. you need to connect that.
The one you have looks like the vent switch.
if it is, it plugs into the back of the intake.
When you switch from defrost to floor does it work?
