Engine: Built TBI 350
Transmission: T-5
Axle/Gears: 3.42 Posi
Is this TBI setup going to work?

I have a 350 with a LT4 hotcam, air gap intake, and vortec heads. Can I put a 454 tbi on without problems? Or am I going to have to chip it?

Oh, it is definitely going to need tuning. And it isn't going to work all that well with an air-gap intake. Wet flow TBI needs some heat in the intake.

A chip and a tune are basically the same thing, but a chip is a physical copy. Where as with tuning software you can get multiple tunes, which is what will be needed. Check out dynamicefi.com

When I dyno it can they tune it?

What did you do with your motor? I see you have a built TBI LO5. How did you tune yours?

If you are going to a tuner who uses a dyno, yes. If your just going to a dyno to put your car on the machine, no.

I went with rbobs EBL Flash. It's not only an amazing piece of tuning software, but it's an outstanding diagnostic system. It gives full read outs of malfunction codes (SES Light) and gives very in-depth readings of ALL sensors. From voltage to steps, counts and lots more!!!! Check out that website or search EBL Flash.

What if it's my daily driver in the summer only? Can I still run an air gap? I'm highly considering getting your EBL flash.

Even in the summer the temperature can vary greatly between early-morning and mid-afternoon. Can try it and see.
